Optimizing Cell Culture Methods for Acetylcholinesterase Production

The Clark Lab currently produces Acetylcholinesterase (AChE) for use in pH-responsive nanosensors in-house, by collecting waste media from transfected HEK293T cells in a medium with 10% Fetal Bovine Serum (FBS). Previous research has shown HEK293T cells to be less efficient in recombinant protein production than other lines such as CHO-K1 cells, and media containing FBS to diminish recombinant protein purity. This project investigates and compares the productivity of cultures following the labs current methods, using transfected CHO-K1 cells, and using HEK293T cells in a serum-free media to find the optimal cell culture methodology for AChE production.
